This project aims to define the molecular events that activate autophagy in response to DNA damage and to determine the scope of autophagy in the degradation and/or sequestration of nuclear proteins. To this end, we will chemically inhibit or knockdown kinases involved in DNA damage signaling and screen for activation of autophagy in a medium throughput manner using a flow cytometry-based assay, thereby identifying and subsequently characterizing critical regulators of autophagy. Further, we will analyze how autophagy affects cell fate upon DNA damage and identify DNA repair factors and chromatin-associated proteins that are targeted by autophagy after DNA damage.
