We observe that the prototypical postsynaptic MAGUK PSD-95 is subject to allosteric regulation. In this study, we will examine the biophysical nature and functional consequences of this protein allostery at synaptic sites. In addition, we will explore if and how such regulation affects other synaptic MAGUKs, focusing initially on MPP2. We will also characterize this new synaptic scaffold protein with regard to its function in the development and maintenance of the postsynaptic architecture, and regarding its role in modulating neurotransmission.
