This central Project administers and performs the services provided by the proteomic platform at the Institute for Biochemistry and Biotechnology of Plants at the University of Münster. In the first and second funding period phosphoproteomic analyses from in vitro and in vivo samples have been established and conducted. The proteomic central project will continue to serve as a platform for peptide and protein identification, analysis of phospho-peptides and ¿proteins and comparative proteomics for the research group. Individual research groups within the Research Unit will perform protein sample preparations prior to proteolytic digestion and mass spectrometric analysis. The central project will have an advisory function in this respect. The proteomic platform will continue to perform proteolytic (tryptic) digestion of the sample and further sample handling, i.e. enrichment of phosphopeptides via SIMAC and ERLIC-TiO2 strategies, and mass spectrometric analyses. The handling and mass spectrometric analyses of in vivo Arabidopsis samples will be in the center of the third funding period. Due to the advent of funding of a new Q-Exactive plus mass spectrometer, a new focus will be put on quantitation of peptide and protein phosphorylation of interest in complex plant samples.
