Reactive metabolites are of key significance for damaging the peritoneal membrane in peritoneal dialysis (PD) which results in ultrafiltration failure. Endothelial aquaporin-1 (AQP1) channels play a central role in this process. Therefore, an endothelial specific AQP1-knockout mouse will be generated and characterized to investigate the physiological relevance of AQP1 in a PD mouse model and in isolated primary endothelial cells. Analysis of human peritoneal biopsies of PD patients will also be performed to elucidate the pathophysiological relevance of AQP1.
