Human hepatitis B (HBV) and delta (HDV) viruses cause acute and chronic hepatitis. Despite a high medical bur-den, early infection events and how episomal viral genomes are maintained are poorly understood. Identification of sodium taurocholate co-transporting polypeptide (NTCP) as receptor and development of HBV/HDV-susceptible cell lines opened a new area in HBV/HDV research. This project aims at visualizing HBV and HDV entry and capsid trafficking applying AAV expression vectors for amber suppression and click labeling. Together with AAV-based perturbation of cellular gene expression, we will dissect the kinetics of attachment and formation of virus-receptor complexes and study intracellular HBV/HDV trafficking, especially nuclear reimport of viral structures.
