We made important progress in identifying relevant viral and cellular factors in plus-strand RNA virus replica-tion organelle (RO) biogenesis and functionality, and revealed surprising parallels between HCV and SARS CoV 2. Furthermore, we identified a sensitive step in Dengue replication that turned out to be well suited for antiviral therapy by employing an integrated mathematical model at single-cell resolution. In the next funding period, we want to continue conducting comparative analyses of viral and host cell factors involved in RO biogenesis and function across phylogenetically distant viruses. Bioinformatics and systems biology approaches will be included to capture the dynamics of RO formation, viral replication and virion assembly.
