In the second period we intend to study the influence of peptide-fused tweezers and artificial cationic peptide receptors on the binding interfaces of the C-terminal domains of UBXD1 and the p97 protomer, on the Taspase-1 loop region as well as on the Survivin dimer interface and its NES region. We already started to optimize NMR studies for the HtrA1 protease and plan to perform titration studies with supramolecular ligands using methyl-TROSY experiments. Additionally, we shall extend side chain directed NMR experiments, which enable proper localization of supramolecular ligands, to higher molecular weight proteins. We also shall investigate the composition of the ligand-shell of nanoparticles and will determine the ligand interaction to proteins.
