Kinetochores are multi-subunit protein assemblies that mediate chromosome segregation during cell division. In this proposal, we will build on our recent in vitro biochemical reconstitution of kinetochores and of their regulation. We will explore the potential of supramolecular compounds generated within the CRC to modulate reversible protein-protein interactions, with a special focus on those mediated by phosphorylation. For this, we plan to use a variety of biochemical assays, including real-time assays that monitor reaction kinetics, which we will use to assess perturbations introduced by supra-molecular ligands.
