Parnafungins belong to a novel class of natural products isolated from fungi. Their biological activity and in particular their action against fungi made led not only to their discovery, but pose also interesting challenges for its total synthesis and delivery of analogues. The stereogenic centers and an unusual and labile isoxazole ring are particular challenges. Based on preliminary results the first total syntheses of these molecules are presented. Key steps are the construction of the xanthone also the connection of the two aromatic building blocks, which were connected via cross-coupling reactions. The xanthone ring will be assembled by established route. Besides the total synthesis, analogues will be prepared and tested in collaboration for their biological activity.
