The polyphosphate/factor XII-driven contact system provides a paradigm to elucidate proinflammatory and procoagulant mechanisms. We aim to analyse functions of the polyphosphate/factor XII-axis in inflammatory, immune, infectious, and malignant liver diseases. Our translational proposal comprises transgenic mice with deficiencies in the polyphosphate/factor XII-pathway and systems biology-based analyses of human liver diseases transcriptomes. The sum of our studies will improve our understanding on the contributions of the polyphosphate/factor XII-driven contact system in liver disease states and its role as potential drug target.
