The B cell antigen receptor (BCR) controls the activation of naïve and memory B cells via common and BCR isotype-specific signaling mechanisms. In project 08, we elucidated several of those mechanisms, showing that the activation of naïve B cells requires the adaptor protein CIN85 whereas memory B cells depend on a BCR-intrinsic signal-amplification motif. Within the next funding period, we will extend this work to a global analysis of signal elements whose subcellular localization is controlled by BCR isotypes and co-receptors. In preliminary experiments, we identified the transcription factor TFEB as a novel B cell nucleocytoplasmic shuttling element, the function of which will be dissected in vivo and in vitro.
