B cell activation is mediated by multiple tyrosine phosphorylation events that are tightly controlled by protein tyrosine phosphatases (PTPs). To better understand the role of different PTPs in hyperactivity and auto-reactivity of B cells, new substrates of the PTPs SHP-1 and PTP1B in B cells were identified by quantitative phosphoproteomic approaches. These PTP substrates will be further studied by knockouts in cell lines and their relevance will be studied in B cells from autoimmune patients. A further emphasis lies on the discovery of new phosphatases in B cell development.
