Our goal is to understand the regulation of gene expression in Drosophila development, and in particular the interplay between transcription factor activity and chromatin organization. Building on a comprehensive genome-wide analysis of core promoters, we plan to use sensitive proteomics approaches to characterize the interactome of selected general transcription machinery components and histone modifications in a promoter-type specific fashion. In a second aim, we seek to characterize the role of pioneer transcription factors in regulating chromatin accessibility at enhancers within the Ecdysone signaling cascade.
