Ex vivo human skin, maintained in short term culture, is the key experimental model of this project, which focuses on drug and nanocarrier penetration. Trans-well penetration assays and dermal microdialysis combined with biomarker analysis (cytokine release, immune cell activation) will allow for preclinical screening of drug delivery and biological effects. A clinical drug study investigating lesional and non-lesional skin of young and aged volunteers with psoriasis is also planned. Joint expertise within SFB 1112 on in vivo assessment methods will be applied to understand characteristics of lesional skin.
