Theoretical investigations on the mechanisms of nanocarrier-based drug transport through skin are pursued, employing a combination of atomistic simulations and stochastic modeling approaches. Multicomponent diffusion models for nanocarrier-assisted drug penetration into skin will be developed. Based on the new methodology developed in the first funding period to extract diffusivity profiles of molecules in lipid membranes from atomistic simulations, the diffusivities of a whole set of drugs in lipid membranes of different composition will be determined. Ultimately, these parameters will be incorporated into a bottom-up model for the transport of drug molecules through the stratum corneum.
