In the past funding period, the endocannabinoid anandamide (AEA) was identified as the most potent endogenous ligand for nuclear receptors of the NR4A family. Now, the project will characterize additional epigenetic mechanisms operative for endocannabinoids and unravel the molecular and structural basis of the AEA-NR4A interaction. Based on the AEA structure, we will develop novel NR4A ligands. In animal experiments, the interaction of NR4A with AEA will be tested as novel anti-inflammatory principle, using conditional knockout mice of NR4A receptors. The newly developed AEA-analogues will be tested in vivo for their therapeutic potential.
