The "Cell-Fate Decision Analysis Platform" supports the projects of the CRC 1054 in their study of cellular determinants of immune cell activation and effector function. Specifically, we have set up the ImageStream Mark II technology for image acquisition and data analysis. This infrastructure now provides support in the detection and quantification of colocalization of proteins of interest with other proteins in the cell or with defined subcellular compartments in specific subpopulations of immune cells. Besides RNA localization in stress granules or P bodies, proximity ligation assays to relate protein interaction to subcellular compartments and observation of signaling molecules in T cell/APC conjugates, we recently expanded the Cell Fate Decision Analysis Platform to include a Cytek Aurora spectral flow cytometer, which allows detection of over 30 colours simultaneously with high sensitivity and resolution. We will support the projects of the CRC 1054 in panel design and computational analysis of high-dimensional flow cytometric data.
