Protein synthesis and degradation are homeostatically coordinated to regulate the neuronal proteome. After characterizing the molecular feedback loop mediating the crosstalk from protein degradation to synthesis, the focus will switch in the third funding period on the converse regulation of protein degradation by protein synthesis. The effects of protein synthesis inhibition on proteasome activation, protein ubiquitination, global protein levels, UPS catalytic activity and local proteasome activity will be assessed in order to fully characterize the homeostatic crosstalk between protein synthesis and degradation.
