This project aims at the design of nanoagents whose operation can be controlled by light. First, we will devise a method to precisely measure temperature distributions and control molecular absorption and desorption on plasmonic nanostructures by a combination of optothermal manipulation and super-resolution microscopy. Secondly, we will investigate how plasmonic DNA-origami nanopores can be incoorporated into lipid bilayer membranes for SERS measurements of single molecules and DNA. Finally, we will explore the use of photolipids as nanoagents to control characteristic membrane properties. In particular, we want to investigate the conditions for two-photon photoisomerization of photoswitchable nanoagents, which is imperative for their application in cellular environments and 3D matrices.
