Project Description

TP12 investigates how the rhomboid protease RHBDL4 resolves and avoids proteotoxic stress in the Endoplasmic Reticulum (ER) and secretory pathway. By combining cell-based studies with proteomics and membrane biochemistry with cryo-electron microscopy, the Lemberg group aims to decipher the molecular mechanism of RHBDL4-catalyzed cleavage. In a second work stream, TP12 will use a combination of tissue culture models and primary cells from RHBDL4 knockout mice to study the function of RHBDL4 in integrating and regulating secretion dynamics in stress response.
