The aim of our study is to understand how different functional states of myeloid cells are linked to oligodendrocytes and remyelination in MS lesions. We will 1. dissect the molecular mechanisms underlying impaired remyelination caused by a maladaptive immune response induced by accumulated myelin debris in myeloid cells and 2. test the hypothesis that lipidated ApoE particles formed after myelin debris clearance are reutilized by oligodendrocytes to provide cholesterol for remyelination. To address our questions we will combine our expertise in oligodendroglial and myelin biology, neuropathology, stem cell technologies and molecular biology.
