Mutations in the Nucleophosmin gene (NPM1mut) are among the most frequent genetic abnormalities in acute myeloid leukemia (AML). The project aims are: 1) Evaluation of the clinical impact of genetic events cooperating with NPM1 mutations in a unique cohort of 588 patients with NPM1-mutant AML from a randomized clinical trial; 2) analysis of clonal genetic evolution patterns using novel single-cell sequencing technologies; 3) functional analyses of cooperating mutations (e.g., of HNRNPK, PTPN11, MYC, RAD21 genes) by using CRISPR/Cas9 technology; 4) study of the impact of NPM1 mutations on the transcriptome and proteome.
