Project Description

To analyze the biology of acute erythroid leukemia (AEL), we will characterize the cellular stage which is permissive for leukemic transformation in our established erythroid leukemia mouse models driven by the homeobox genes Cdx4 and VENTX. We will perform a comprehensive characterization of the transcriptome, methylome and proteome of human AEL, enriched by analysis of signaling pathways at the single-cell level using mass cytometry. In addition, we will analyze the functional relevance of potential collaborating mutations such as TP53, NPM1, DNMT3A and ASXL1 mutations using CRISPR/Cas9 technology.
