Final Report Abstract

The results of this project provided new insights into the transcriptional activity of the rye B chromosome and changed the prevalent view that B chromosomes (Bs) are silent genetic elements. We found that B chromosomes similar to A chromosomes (As) are interacting with active form of RNA polymerase II. We showed that genic sequences located on B chromosomes are transcriptional active. In addition, the transcriptional activity of the rye B was shown to be regulated in different tissues and with ability to modulate the transcriptional activity of A chromosomes in trans. Sequences comparison revealed that Bs have accumulated mutation in contrast to A-located genic sequences. To investigate the influence of B chromosomes on the transcriptome profile of rye and also to determine B-specific transcripts and those A-located genes which are differentially expressed in presence of Bs, RNAseq was performed using polyadenylated RNA from vegetative (leaf and root) and generative (anther) tissues of plants with and without Bs. We could show that presence of Bs increase transcriptional activity of repeat elements especially in generative tissues.
