The human ABC transporter MDR3 catalyses the flop of PC-lipids from the inner to the outer leaflet of the hepatocyte membrane. Together with BSEP, which transports bile acids (BA) and ABCG5/G8 translocating cholesterol, mixed micelles in the canalicular duct. Based on the results of the second funding period, we will continue the studies aimed to unravel the mechanism of PC-lipid transport. Therefore, we will establish an in vitro liposomal-based assay for detailed functional analysis. In parallel, we will analyse the interplay of MDR3 with BSEP or BSEP and ABCG5/G8 in cell culture and the modulation of this transporter network by BA, lipids and/or small molecules.
