The project aims at clarifying the composition and regulation of multiprotein complexes. In particular, we employ novel microscopic and functional assays to unravel the role of protein phosphorylation and its regulatory impact on protein-protein interactions at integrin-rich focal adhesion sites. Together with collaboration partners within the CRC we exploit genetic and chemical-biological screens as well as chemical modification of small molecule compounds to directly modulate the activity of protein kinases and phosphatases, which we have identified to be involved in the assembly and disassembly of focal adhesion sites. These approaches will afford us not only with detailed functional insight, but will also open up novel avenues to study phosphorylation-dependent control mechanisms orchestrating multiprotein complexes.
