Organization of the synaptic active zone scaffold by Bruchpilot and RIM-binding proteinNeurotransmission at chemical synapses depends on elaborate protein scaffolds that decorate active zones. In project A06, we aim at devising a pseudo-atomic model of the Bruchpilot/RIM-binding protein-based core of the active zone scaffold at Drosophila neuromuscular junctions. To this end, we will combine results from protein crystallography, biochemical structure probing, phylogenetic co-variation analysis, super-resolution light microscopy and cryo-electron tomography in an integrative structural modeling approach.
