BamA in bacteria and Tob55 in mitochondria are the central components in complexes that mediate assembly of ß-barrel proteins. Both proteins contain POTRA domains with an unclear function. To better understand the role of these domains we will construct hybrid Tob55/BamA proteins with differently configured POTRA domains and will examine the activity of such hybrid proteins in both mitochondria and bacteria. In the second part of the project we will compare the assembly of ß-barrel proteins to that of secretins. Using yeast as a model system we will dissect the stages in the assembly process of secretins and investigate the requirement for periplasmic assembly factors.
