Interference with senescence-dependent mechanisms to improve renal transplant outcome (C08)

Project C08 studies renal senescence as a cause of chronic graft dysfunction after kidney transplantation. The project has demonstrated that lesions, which characterize a chronic graft dysfunction like interstitial fibrosis and tubular atrophy, also show telomere shortening and up-regulation of the senescence-associated cell cycle inhibitor p16INK4A. In the next funding period, C8 They will test whether removal of senescent cells will enhance the regeneration of kidney grafts of older donors. In a second approach, they will test if silencing of p16INK4A will rescue defective regeneration caused by dysfunctional telomeres. In a third approach, they will test if cyclin D1 also contributes to the ageing phenotype and defective repair mechanisms. Results from C8 will be important to overcome the problems of chronic graft dysfunction and to enable transplantation of organs from older donors.
