Infectious diseases caused by a prolonged phase of immunodeficiency represents another major clinical problem, which causes morbidity and mortality after hematopoietic stem cell transplantation. Project A7 is searching for possibilities for a faster T-lineage reconstitution. To address this topic, they have investigated markers responsible for directing lineage decision into the T-cell lineages. To this end, they identified several miRNAs with substantial effects on T-cell regeneration. They now aim to identify the relevant target genes using the CRISP/Cas9 technology. In a translational approach, they will use the same CRISPR/Cas9 system to modify target genes in human pre-T-cells.
