Ongoing and planned research projects in the main applicant‘s laboratory are centered on the function of Ca2+ in plant signaling and development. Moreover, we are currently advancing modern cell biological techniques for in planta protein-protein interaction analyses and FRET-based or ratiometric analyses for cellular Ca2+ and ROS measurements that rely on the use of fluorescent reporter proteins and require state of the art confocal microscopy. All these approaches rely on the use of a confocal microscope that is able to perform high-speed multi-channel imaging at highest possible resolution. Similarly, the research of the four laboratories that contribute to this application depends on CLSM equipment that allows for multi-color high-speed imaging and FRET-based Ca2+ analysis. All existing confocal microscopes in the department are either not suitable for such analysis or their intensive use prevents their application in the described research projects. Moreover, the requested CLSM will expand the currently absolutely limited technical capacity to perform especially long term confocal microscopy experiments that currently represents a severe bottleneck especially for the groups that perform research on algae, higher plants or fungi.
