Project Description

The cytokine interleukin(IL)-1 regulates inflammatory cell functions through a largely unexplored nuclear signaling network. In the preceding funding period we found evidence for (i) IL-1-specific enhancers, (ii) novel components that are assembled at IL-1-regulated regions within chromatin and (iii) cytokine-mediated changes in chromatin structure. We plan to reveal the underlying mechanisms by genome-editing and to test the concept of inflammatory transcription factories. Identification of such nuclear control points will open new options to modulate the biological activity of IL-1 in disease.
