The medial septum has an essential role for theta rhythm generation in the hippocampus and is critical for learning and memory. In this proposal we will address the question whether HCN channels in the medial septum are important for this rhythm. We will specifically knock out HCN2 in distinct subsets of medial septal neurons (global, GABAergic and glutamatergic) in the mouse. We will investigate how loss of HCN2 alters firing pattern in medial septal neurons in acute brain slices (aim #1) and in intact septo-hippocampal preparation (aim #2). In aim #3 we will characterize in vivo function of HCN2 using depth recording in anesthesized mice, EEG telemetry and behavioral experiments.
