The project focuses on the calpain family of Ca2+-activated proteases and their endogenous inhibitor protein calpastatin (CAST) as well as on the role of the calpain/CAST proteins in processes usually attributed to redox signalling. Particular attention will be paid to regulation by microRNA and the consequences of calpain activation/inactivation on the mitochondrial proteome and function, including alterations in -ketoglutarate levels and the activity of members of the Jumonji C domain-containing histone lysine demethylase (Jmj-KDM) family.
